The College of Pharmacy discussed the PhD dissertation entitled \u201cThe Improving Effects of Different Doses of L-Carvone in Male Mice with Glycerol-Induced Renal Injury Through Rhabdomyolysis-Mediated Effects\u201d, by the student Abeer Riyadh Abdali and the supervisor, Professor Dr. Sarmed Hashim Kadhim, at the Pharmacology and Toxicology Department.<\/span><\/p>\n

The study aimed to investigate the effects of different doses of L-Carvone on male mice with glycerol-induced nephropathy via rhabdomyolysis-mediated mechanisms.<\/span><\/p>\n

The study included fifty male mice randomly divided into five groups of 10 and treated as follows: Group I mice received corn oil orally at a dose of 1 mg\/kg for five consecutive days and an intramuscular (IM) injection of normal saline at a dose of 10 mg\/kg on day five; Group II mice received an IM injection of 50% glycerol at a dose of 10 mg\/kg; Group III mice received L-carvone orally at a dose of 25 mg\/kg for five consecutive days and an IM injection of 50% glycerol at a dose of 10 mg\/kg on day five; Group IV mice received L-carvone orally at a dose of 50 mg\/kg for five consecutive days and an IM injection of 50% glycerol at a dose of 10 mg\/kg on day five; Group V mice received L-carvone orally at a dose of 100 mg\/kg for five consecutive days and an IM injection of 50% glycerol at a dose of 10 mg\/kg on day five. On day six, the mice were sacrificed, and blood and kidney tissue samples were collected for biochemical, gene expression, and histological analysis.<\/span><\/p>\n

The results showed that treatment with L-carvone significantly reduced the histological changes induced by glycerol in renal tissue and lowered serum levels of BUN, creatinine, myoglobin, and CK-MM, as well as KIM-1 and NGAL levels in renal tissue samples. Furthermore, pretreatment with L-carvone alleviated glycerol-induced inflammation and apoptosis by downregulating the expression of TNF-\u03b1, IL-1\u03b2, and NF-\u03baB, reducing the levels of BAX, caspase-8, and caspase-3, and increasing Bcl-2 levels in renal tissues.<\/span><\/p>\n

The study recommended conducting further research using different doses of L-carvone over longer periods to evaluate the protective efficacy of this compound against nephrotoxicity and to investigate its molecular mechanisms in greater depth.
