The College of Pharmacy discussed the PhD dissertation entitled \u201cAssociation of Lysyl Oxidase Gene” Variations (rs1800449) with Serum Biomarkers and Breast Cancer Development in a Sample of Iraqi Women\u201d by the student Shahad Sabah Khalid and the supervisor, Professor Dr. Shatha Hussein Ali, and Dr. Ahmed Zahair Alsammarraie, at the Clinical Laboratory Science Department.<\/span><\/p>\n

The study aimed to investigate the presence of the single nucleotide polymorphism (SNP) at the G473A locus of the lysyl oxidase (LOX) gene (rs1800449) and to evaluate its association with the development of breast cancer among Iraqi women. Determine the potential relationship between this polymorphism and the serum levels of lysyl oxidase (LOX), fibronectin-1 (FN-1), tumor necrosis factor-alpha (TNF-\u03b1), transforming growth factor-beta 1 (TGF-\u03b21), homocysteine (Hcy), hypoxia-inducible factor-1 alpha (HIF-1\u03b1). Furthermore, the study evaluated the potential role of serum lysyl oxidase as a diagnostic biomarker for the detection and progression of breast cancer.<\/span><\/p>\n

The study included 90 Iraqi women aged between 30 and 70 years. Blood samples were analyzed to determine serum levels of TNF-\u03b1, Hcy, LOX, HIF-1\u03b1, cancer antigen 15-3 (CA15-3), fibronectin (FN), and TGF-\u03b21 using specialized enzyme-linked immunosorbent assay (ELISA) kits. Genomic DNA was extracted from peripheral blood leukocytes using a commercial DNA extraction kit and stored at \u221220\u00b0C for subsequent genotyping of the G473A single nucleotide polymorphism (rs1800449) in the LOX gene.<\/span><\/p>\n

The results showed that the LOX gene polymorphism (rs1800449, C\/T) was not significantly associated with breast cancer susceptibility, despite a higher frequency of the CT and TT genotypes among patients. The CC and CT genotypes were significantly associated with elevated serum biomarker levels compared with the control group. Significant differences in serum HIF-1\u03b1, CA15-3, and TGF-\u03b21 levels were observed among different genotypes in breast cancer patients. A moderate positive correlation was found between HIF-1\u03b1 and CA15-3, whereas weak correlations were observed between Hcy and LOX, and between HIF-1\u03b1 and FN. TNF-\u03b1, Hcy, CA15-3, and FN demonstrated excellent diagnostic performance, while LOX, HIF-1\u03b1, and TGF-\u03b21 showed high sensitivity and specificity, supporting their potential value as promising biomarkers for breast cancer diagnosis.<\/span><\/p>\n

The study recommended conducting larger multicenter studies involving larger sample sizes to further investigate the association between the LOX gene polymorphism (rs1800449) and breast cancer susceptibility across different populations. It recommended future studies to evaluate changes in serum biomarker levels according to disease stage and treatment response while correlating these findings with the clinical and molecular characteristics of the tumor. Furthermore, further investigation of lysyl oxidase as a potential diagnostic biomarker and therapeutic target, particularly in aggressive breast cancer subtypes, is recommended.<\/span><\/p>\n
