The Faculty of Pharmacy discussed the Master’s Degree thesis, tagged “Hepatoprotective effect of triple omega 3-6-9 against acute acetaminophen-induced hepatic injury as compared with the effect of N-acetylcysteine in mice.” by Yousif Hashim Mohammed and his supervisor, Assist. Prof. Dr. Ali Faris Hassan in the Department of Pharmacology and Toxicology on July, 23, 2023. This study was designed to investigate the possible hepatoprotective effect of triple omega 3-6-9 against acute acetaminophen-induced hepatic injury as compared with the effect of N-acetylcysteine in mice by evaluating liver function enzymes and some selected inflammatory markers in rats’ serum, in addition to rats’ brain tissue histopathology. The results showed that the mice in Group 2 (positive control) after a single intraperitoneal injection with acetaminophen produced a significant increase in the serum levels of tumor necrosis factor alpha, interleukin 10, interleukin 1b, alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase and produced a significant decrease in the reduced glutathione, glutathione peroxidase, and superoxide dismutase in the liver, with a significantly increased hepatic malondialdehyde level as compared with Group 1 (negative control). The mice in Group 3 (treated with triple omega 3-6-9 at dose 50 mg/kg ) ,Group 4 (treated with triple omega 3-6-9 at dose 100 mg/kg) and Group 5 (treated with N-acetylcysteine (100 mg/kg) prior to acetaminophen produced a significant decrease in the serum levels of tumor necrosis factor alpha , interleukin 10, interleukin 1b, alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase and produced a significant increase in the reduced glutathione, glutathione peroxidase, and superoxide dismutase in the liver with a significantly decreased hepatic malondialdehyde level as compared with Group 2 (positive control) , while mice in Group 5 as compared with Group 1 produced a significant different in level of aspartate aminotransferase, interleukin 10, interleukin 1b, superoxide dismutase , glutathione, glutathione peroxidase. In comparison to Group 2,observable characteristics of liver sections of the pretreatment groups with triple omega 3-6-9 and N-acetylcysteine exhibited significant improvement, as determined by the findings of the histological inspection. Overall, this study clearly showed the administration of triple omega 3-6-9 to mice led to a reduction in the toxicity of acetaminophen. These results indicate that triple omega 3-6-9 provides protection against the toxicity that is produced by acetaminophen in the liver. One possible explanation for this is that it possesses anti-inflammatory and antioxidant effects.
